This proposed study will look to elucidate the role of adhesion complexes in a bacterial response to stress. Gram-positive bacteria of the genus Streptococcus utilize adhesins to adhere to human oral epithelial cells. The Gram-positive Streptococcus mutans is one of the leading causes of caries in humans. When this bacterium lives in the mouth it is exposed to multiple stresses which include: acidic environments, low pH, and high temperatures. These conditions can be quite damaging to bacterial cells. Streptococcus mutans has a unique lantibiotic system which has been proposed to serve as an additional protective mechanism against these stresses. Streptococcal cells produce a bactericidal effector protein, Streptococcal Zinc-metalloprotease (SZP), which is secreted into the environment. SZP is predicted to kill Gram-positive bacteria by cleaving the polymer of glucans which comprise the cell wall. SZP has also been shown to have proteolytic activity on proteins and peptides. It has been speculated that SZP may act as a cell-to-cell signaling agent which regulates adherence and may be a component of a bacterial response to stress. This study will investigate the role of these adhesion complexes in SZP mediated-bacterial killing by determining how cell-to-cell signaling is accomplished, and how SZP interacts with these adhesion complexes. This study will also determine if adhesion complexes are required for the ability of SZP to kill Gram-positive bacteria in a stress free environment. The results of this study will help us understand the role of adhesion complexes in bacterial adhesion and signaling in addition to their role in bacterial killing.![](br
